The Portuguese side had mistakenly included the Belgian striker in the squad list for the two-legged tie.
However, as Depoitre already featured for Gent in the third preliminary round of the Champions League against Viitorul Costanza, he is ineligible.
Porto do have the opportunity to replace him with a different player within 24 hours of the first leg match.
